Title:
Application of single-cell genomics to dissect tissue heterogeneity in development and disease
Abstract:
Tumor heterogeneity is thought to be one of the major contributing factors to therapy failure and cancer progression. However, conventional methods for profiling molecular content of tumors often characterize them at the population level. Recent advances in single-cell genomics provide a way to overcome this hurdle. Several years ago, we adopted a single-cell RNA sequencing platform (Drop-seq) to analyze transcriptional profiles of thousands of individual cells simultaneously. I will describe our efforts to map individual cells in the Drosophila eye during normal development and upon inactivation of the Retinoblastoma (RB) tumor suppressor pathway. I will illustrate how we employ single-cell data as an entry point for the follow up genetic and cell biology experiments to dissect complex mutant phenotypes.
